This study focuses on optimizing a protocol to derive astroglial cells from induced pluripotent stem cells in order to model neuroinflammation in spinocerebellar ataxia type 17. The researchers evaluated different culture protocols to analyze astrocyte gene expression profiles and assessed NF-kappaB signaling path activity in cells derived from healthy donors compared to those from affected patients. The results showed that the FHIA protocol yields the expression profile closest to mature astrocytes, while a protocol utilizing one percent FBS generates the least differentiated astroglial cells, making it suitable for studying early neuroinflammatory mechanisms. Furthermore, astrocytes derived from spinocerebellar ataxia type 17 patients exhibited higher baseline NF-kappaB pathway activation than control cells, pointing toward a pre-existing pro-inflammatory status in patient astrocytes.
